A Safer Opioid Painkiller with Low Side-Effects

The Ministry of Science and Technology has presented its 2020 Future Tech Breakthrough Award (Heterocyclic compounds and use thereof) to the National Health Research Institutes (NHRI) for the discovery of painkilling compounds that are safer and have fewer side effects than morphine. The work was performed by Dr. Shau-Hua Ueng’s group and Dr. Shiu-Hwa Yeh’s group. The series of compounds they found demonstrates better antinociception with fewer adverse effects than morphine.

Morphine is widely used for the treatment of severe pain. However, clinical usage of morphine also results in tolerance, dependence, constipation, sedation, heart function depression, and respiratory depression. Patients who need pain control suffer from these clinical adverse effects caused by morphine. Dr. Shau-Hua Ueng and Dr. Shiu-Hwa Yeh discovered a series of opioid receptor/nociceptin receptor dual agonists that demonstrated fast and potent antinociception, and impressively fewer side-effects than morphine in dependence, constipation, heart function depression, and respiratory depression. These dual agonists are patentable and have the potential to lead to a safer pain killer. This breakthrough might provide more comfortable pain control to patients after surgery and to those suffering from cancer pain.

The findings of the most recent study were published in Neuropharmacology in April 2020.
